CD226 (DNAM-1) is a transmembrane glycoprotein expressed predominantly on immune cells like T cells, NK cells, and monocytes. It contains two immunoglobulin V-like domains (D1 and D2) in its extracellular region, a type I transmembrane domain, and an intracellular cytoplasmic domain with critical phosphorylation sites at Y322 and S329 in humans.
