CITED2 is a highly conserved transcriptional co-regulator that lacks a DNA-binding domain and functions by interacting directly with a broad range of transcription factors and co-factors, including TFAP2, SMAD2/3, PPARγ, oestrogen receptor, MYC, Nucleolin, and the co-activators p300/CBP. It is characterized by a conserved C-terminal domain (CR2), which is crucial for its transcriptional regulatory activity and enables high-affinity interaction with the CH1 domain of p300/CBP. Depending on the literature source, CITED2 may also be discussed as MRG1 and Cbp/p300-interacting transactivator 2.
