Eps15 (EGFR pathway substrate 15) was originally identified as a substrate for the kinase activity of the EGFR. It has a tripartite structure comprising an NH2-terminal portion with three EH domains, a central putative coiled-coil region, and a COOH-terminal domain with multiple Aspartate-Proline-Phenylalanine triplets. Eps15 localizes at clathrin-coated pits, interacting with the clathrin assembly complex AP-2 and the AP-2 binding protein Epsin.
