About the Target

Fibroblast growth factor 21 (FGF21) is a crucial endocrine regulator primarily produced in the liver, adipose tissues, and skeletal muscles. It plays a significant role in metabolic homeostasis by enhancing hepatic insulin sensitivity, reducing lipogenesis, and promoting fatty acid β-oxidation, thereby mitigating liver diseases like non-alcoholic fatty liver disease (NAFLD) and alcoholic fatty liver disease (AFLD).

Reported cellular context includes secreted, which can matter when signal is compared across treatments or changing cell states. Following FGF21 across matched perturbations can help separate abundance effects from shifts in localization, complex assembly, or pathway state.

Research Context

FGF21 is commonly interpreted in the context of metabolism research, and readouts are often stronger when a study separates expression changes from compartment-level redistribution. When reported signal spans secreted, a defined reference condition can make comparisons more interpretable across perturbations, passages, or replicate sets.

Consider these angles when interpreting target-level changes:

  • signal enrichment within secreted relative to the broader cellular background
  • responses linked to nutrient status, mitochondrial state, or metabolic rewiring
  • co-patterning with orthogonal markers and control conditions that clarify pathway state
  • time-matched comparisons so changes reflect biology rather than handling or sampling drift

Variant Considerations

If your project spans exploratory questions, the regular version offers a balanced option for establishing baseline signal behavior for FGF21. This can help when protocols evolve over time and the goal is to compare experiments using a stable reference workflow.

Standardize sampling time, control choice, and downstream analysis thresholds so apparent differences in FGF21 reflect biology rather than handling. When interpreting FGF21, it is often useful to decide early whether the main question is overall abundance, compartmental enrichment, or context-dependent redistribution.

For multi-run studies, a shared reference condition can keep FGF21 trends easier to compare across datasets. That kind of consistency is especially helpful when follow-up work expands to new perturbations, model systems, or longitudinal collections.

Targets:
FGF21
Research Area:

Application:
IHC • WB
Reactivity:
Human • Mouse • Rat
Specificity:
FGF21 Antibody [H23F24] recognizes endogenous levels of total FGF21 protein. The expression level of FGF21 varies in different sample types (PMID: 27285327). For cell lines, the culture supernatant must be collected for detection at the same time, because the target protein is easily secreted into the external environment (PMID: 24041694). FGF21 Rabbit mAb is not recommended for Mouse-WB and Human-lHC (Paraffin).
