About the Target

Matrin 3 (MATR3) is a highly conserved, ubiquitously expressed inner nuclear matrix protein that serves as a scaffold for chromatin organization and nuclear architecture. The 847-amino acid protein features two C2H2-type zinc finger domains (ZF1: residues 288-322, ZF2: 798-833) for DNA binding, two RNA recognition motifs (RRM1: 398-473, RRM2: 496-575) for RNA interaction, and a nuclear localization signal (588-595), with the remainder of the protein comprising largely intrinsically disordered regions, especially acidic at the C-terminus. Depending on the literature source, MATR3 may also be discussed as Matrin 3 and KIAA0723.

Reported cellular context includes nucleus, which can matter when signal is compared across treatments or changing cell states. Following MATR3 across matched perturbations can help separate abundance effects from shifts in localization, complex assembly, or pathway state.

Research Context

MATR3 is commonly interpreted in the context of metabolism and developmental biology research, and readouts are often stronger when a study separates expression changes from compartment-level redistribution. When reported signal spans nucleus, a defined reference condition can make comparisons more interpretable across perturbations, passages, or replicate sets.

Consider these angles when interpreting target-level changes:

  • signal enrichment within nucleus relative to the broader cellular background
  • responses linked to nutrient status, mitochondrial state, or metabolic rewiring
  • stage-dependent patterns during differentiation, morphogenesis, or lineage commitment
  • co-patterning with orthogonal markers and control conditions that clarify pathway state

Variant Considerations

If your project spans exploratory questions, the regular version offers a balanced option for establishing baseline signal behavior for MATR3. This can help when protocols evolve over time and the goal is to compare experiments using a stable reference workflow.

Standardize sampling time, control choice, and downstream analysis thresholds so apparent differences in MATR3 reflect biology rather than handling. When interpreting MATR3, it is often useful to decide early whether the main question is overall abundance, compartmental enrichment, or context-dependent redistribution.

For multi-run studies, a shared reference condition can keep MATR3 trends easier to compare across datasets. That kind of consistency is especially helpful when follow-up work expands to new perturbations, model systems, or longitudinal collections.
