MKK3 and MKK6 are closely related dual-specificity protein kinases responsible for activating p38 MAP kinase. They share the function of phosphorylating and activating p38 MAP kinase at its activation site, Thr-Gly-Tyr, while they do not affect Erk1/2 or SAPK/JNK. This phosphorylation of p38 MAP kinase significantly enhances its ability to phosphorylate protein substrates like ATF-2 and Elk-1. Depending on the literature source, MKK3 may also be discussed as MEK3.
