6-Phosphofructo-2-kinase/fructose-2,6-bisphosphatase (PFK-2/FBPase) is an enzyme that both synthesizes and degrades fructose 2,6-bisphosphate (F2,6BP), a potent activator of 6-phosphofructo-1-kinase. Fructose 2,6-bisphosphate enhances the activity of phosphofructokinase, a key rate-limiting enzyme in glycolysis, through allosteric regulation. There are four distinct PFKFB isoforms: PFKFB1, PFKFB2, PFKFB3, and PFKFB4, each encoded by separate genes (PFKFB1-4).
