SMYD2 (SET and MYND domain-containing protein 2) is a protein methyltransferase that methylates histone H3 at lysines 4 (H3K4) and 36 (H3K36), as well as various nonhistone proteins, influencing transcriptional regulation and post-translational modification crosstalk. Structurally, SMYD2 features five domains-S-sequence, MYND domain, insertion SET domain, cysteine-rich post-SET domain, and tetratricopeptide repeat (TPR) domain-forming a bilobal structure with an autoinhibited conformation regulated by the C-terminal domain.
