About the Target

USP39 (Ubiquitin-Specific Peptidase 39) is a member of the ubiquitin-specific protease (USP) family that regulates protein stability by removing ubiquitin chains from substrates. USP39 lacks full catalytic activity because it does not contain all the critical residues in its catalytic triad. Its structure includes a zinc finger-like ubiquitin-binding domain and RNA-binding motifs, which are involved in pre-mRNA splicing and spliceosome assembly. Depending on the literature source, USP39 may also be discussed as CGI-21 and HSPC332.

Reported cellular context includes nucleus and spliceosome, which can matter when signal is compared across treatments or changing cell states. Following USP39 across matched perturbations can help separate abundance effects from shifts in localization, complex assembly, or pathway state.

Research Context

USP39 is commonly interpreted in the context of cancer, cell cycle, and cell signaling research, and readouts are often stronger when a study separates expression changes from compartment-level redistribution. When reported signal spans nucleus and spliceosome, a defined reference condition can make comparisons more interpretable across perturbations, passages, or replicate sets.

Consider these angles when interpreting target-level changes:

  • apparent redistribution between nucleus and spliceosome across matched conditions
  • changes associated with proliferative state, oncogenic signaling, or treatment response
  • cell-cycle linked differences in abundance, timing, or compartmental enrichment
  • signal-dependent shifts after ligand, inhibitor, or growth-factor perturbation

Variant Considerations

If your project spans exploratory questions, the regular version offers a balanced option for establishing baseline signal behavior for USP39. This can help when protocols evolve over time and the goal is to compare experiments using a stable reference workflow.

Standardize sampling time, control choice, and downstream analysis thresholds so apparent differences in USP39 reflect biology rather than handling. When interpreting USP39, it is often useful to decide early whether the main question is overall abundance, compartmental enrichment, or context-dependent redistribution.

For multi-run studies, a shared reference condition can keep USP39 trends easier to compare across datasets. That kind of consistency is especially helpful when follow-up work expands to new perturbations, model systems, or longitudinal collections.
